Decrease of PARP3 and increase in telomerase activity in Saos-2 cell line In the cell line Saos-2 we initially developed an approach similar to that described for the A549 line. Thus, in order to characterize this cell line we evaluated PARP3 mRNA levels by qRT-PCR, and analyzed telomerase activity. Results revealed low levels of enzyme activity. Following, we performed experiments aimed at silencing PARP3 in this cell line, then checking whether this silencing led to an increase in telomerase activity in cells. shRNA-mediated gene silencing allowed us to select the clone of Saos-2 cells with the highest reduction of PARP3, whose mRNA levels decreased by 60% with respect to the control, as qRT-PCR assays showed (Figure 5A).
